The objectives of this doctoral dissertation were to determine thechemical composition of commercial basil, sage, and winter savoryessential oils, to establish their antimicrobial and antivirulent potentialsas well as their ability to enhance the activity of the antibioticciprofloxacin in the controlling the growth of Pseudomonas aeruginosaand Klebsiella spp. clinical isolates. These species represent the mostcommon pathogenic strains characterized by increased resistance andare leading causes of chronic infections due to their ability to producebiofilm.The chemical analysis of the oils revealed a high prevalence of themonoterpene class in all three oils and a high content of the componentsα-thujone, camphor, linalool, (E)-anethole, thymol and p-cymene.After their application to P. aeruginosa isolates, basil and sage oilsshowed antimicrobial activity in the range of 5-20 mg/ml, while wintersavory oil activity ranged from 0.63 to 10 mg/ml. When applied toKlebsiella spp. isolates, the activity range of the basil oil was 2.5-10mg/ml, 1.25-5 mg/ml for winter savory oil, while sage oil was activeonly in concentrations higer than 20 mg/ml. The results indicate thereducing potential of the tested oils in all tested biofilm formationstages, and that they completely degrade the mature biofilm in P.aeruginosa isolates, while in Klebsiella spp. that effect is somewhatlower. The results from the tests investigating the activities of the oilson other P. aeruginosa virulence factors showed that all three oilssignificantly reduce pyocyanin production, while basil and sage oilsshowed abilities to reduce motility in P. aeruginosa isolates, which wasnot observed after the application of winter savory oil. The tested oilsalso led to increased activity of the antibiotic ciprofloxacin afterapplication to the mentioned clinical isolates.
